Will Halo 3 feature proximity voice and teamspeak features the way Halo 2 does?

Again, Bungie has said nothing yet, but here's hoping they change things. Speech in Halo 2, in my opinion, doesn't work terribly well. Enemies can always hear you within a given radius, even through structures, while the interface requires you to specifically signal that you want to communicate to remote teammates-- and even when you do so, nearby enemies can still overhear you.

What difficulty settings will H3 feature?

Bungie hasn't said so but it certainly seems likely that they would stick with the Easy, Normal, Heroic and Legendary modes from Halo 1 and Halo 2. UPDATE: Whether Halo 3 will have any difficulty-modifying skulls, such as the Mythic and Thunderstorm skulls in Halo 2, is unknown. Although most skulls have some effect on difficulty, those two in particular do so, as Thunderstorm changes all enemies to specops, and some players refer to "Mythic" as a separate difficulty level, even though it is not accessible from the menu as such.

Will H3 weapons spawn the way they did in H1 (timed), or the way they did in H2 (when dropped)?

It's most likely way too early to answer this, although presumably Bungie is well aware of the concerns of fans regarding both methods. UPDATE: With a public beta test scheduled for spring 2007, it's likely many fans of the game will have a chance to test what spawn methods Bungie has implemented and to offer their opinion.

Will Halo 3 feature network cooperative play?

UPDATE: The below was written in early 2007. Approximately two months before release, Bungie announced that Halo 3 will feature cooperative play via split screen (two players per box) as well as system link and Xbox Live (four players per game). In a four-player game, the host players the Master Chief, and the other players are the Arbiter and two other Elite warriors.

Any combination can be used; four players can use separate consoles over XBL or system link; two players can share a box and connect to players on one or two other separate boxes.

Are the Cortana Letters canon again, now that Cortana has quoted T.S. Eliot in the H3 trailer?

I doubt Bungie will address this directly, but I don't think so. Clearly some similar themes are being explored, and Bungie may very well be giving a tip of the hat to old school fans with this reference. However, the Cortana in the Cortana Letters is still quite clearly a different character from the Cortana we know in the shipping Halo games, and it seems doubtful to me that further analysis of the Cortana Letters, as entertaining as it is, will prove useful or relevant to the events of Halo 3.

Will Halo 3 include other playable characters besides the Chief?

UPDATE: The below was written in early 2007. Approximately two months before release, Bungie announced that Halo 3 will feature cooperative play via split screen (two players per box) as well as system link and Xbox Live (four players per game). In a four-player game, the host players the Master Chief, and the other players are the Arbiter and two other Elite warriors.

Will Halo 3 include other Spartans?

UPDATE: The below was written early in 2007. By mid-summer 2007, most of Halo 3's feature list was known; to date there is no mention of other Spartans, and the four-player cooperative feature uses the Chief, the Arbiter, and two Elite warriors as characters.

This is a perennial favorite question ever since the Halo backstory in Fall of Reach revealed that other Spartans did, in fact, survive.

Bungie so far has not announced anything specific regards to plot or gameplay features.

What platforms will Halo 3 run on?

Bungie has only announced it for the Xbox 360. It is so extremely unlikely that it will be backported to the original Xbox that I am inclined to call this an impossibility. I doubt the original Xbox's hardware could handle the game at all, and I doubt Microsoft, Bungie, or anyone else besides someone determined not to buy an Xbox 360 would be pleased by a version of the game that could.

Is Halo 3 the last, final, ultimate Halo game ever?

Bungie addresses this in their own FAQ but is a tiny bit ambiguous. The announcement press release seems to indicate yes; their FAQ equivocates, saying that Halo 3 concludes the current story arc (pun most likely intended; looks like Bungie finally hired Stuntmutt). There is every indication that Bungie has been undergoing fatigue over the Halo franchise since crunch time on Halo 2, and I would be very surprised if Bungie did not want to work on something entirely different after Halo 3, or if indeed Bungie has a second team quietly working on something else right now.
